چکیده

The public is increasingly exposed to various engineered nanomaterials because of their mass production and wide application. Even when the biological effects of nanomaterials have been assessed, the underlying mechanisms of action in vivo are poorly understood. The present study was designed to seek a simple, effective, and oxidative stress-based biomarker system used for screening toxicity of nanomaterials. Nano-ferroso-ferric oxide (nano-Fe3O4), nano-silicon dioxide (nano-SiO2), and single-walled carbon nanotubes (SWCNTs) were dispersed in corn oil and characterized using transmission electron microscopy (TEM). Rats were exposed to the three nanomaterials by intratracheal instillation once every 2 days for 5 weeks. We investigated their lung oxidative and inflammatory damage by bronchoalveolar lavage fluid (BALF) detection and comparative proteomics by lung tissue. Two-dimensional electrophoresis (2-DE) of proteins isolated from the lung tissue, followed by matrix-assisted laser desorption-ionization time-of-flight mass spectrometry, was performed. In the present study, we chose to detect lactate dehydrogenase, total antioxidant capacity, superoxide dismutase, and malondialdehyde as the biomarker system for screening the oxidative stress of nanomaterials and IL-6 as the inflammatory biomarker in BALF. Proteomics analysis revealed 17 differentially expressed proteins compared with the control group: nine were upregulated and eight were downregulated. Our results indicated that exposure by intratracheal instillation to any of the three typical nanomaterials may cause lung damage through oxidative damage and/or an inflammatory reaction.

Investigating the protective effects of aqueous extract of the wormwood plant (Artemisia absinthium) on alumina nanoparticle-induced pulmonary toxicity in male Wistar rats

Introduction: Although aluminum oxide nanoparticles (Al2O3-NPs) are the most widely used nanomaterials, limited studies have been reported on their toxicology. Therefore, the present study aimed to investigate the potential toxicity of aluminum oxide (alumina) nanoparticles and the protective role of aqueous extract of wormwood plant on nanomaterial-induced disorders in the lung of rats. Mater...

Dose-Dependent inhibitory Effect of Ferulic Acid, a Dietary Antioxidant on Nicotine-Induced Tissue Oxidative Stress in Experimental Rats

The present study was aimed at elucidating the protective effect of ferulic acid (FA), a natural polyphenol against nicotine-induced tissue damage, including lung, liver and kidney of experimental rats. Female albino rats of Wistar stain were used for the experimental study. Lung toxicity was induced by subcutaneous injection of nicotine at a dose of 2.5 mg/kg body weight (5 days a week, for 22...

Study of Serum and Tissues Angiotensin Converting Enzyme (ACE) Activity in Rat with Gentamicin Induced Renal Toxicity

The angiotensin I-converting enzyme (ACE) converts the inactive angiotensin I molecule to the active angiotensin II. ACE is rich in epithelium, endothelium, and neuroepithelial cells and it found largely on the brush border of intestine and kidney proximal tubules. ACE also presents in the serum. Some pulmonary and renal toxic drugs change the serum and tissue ACE contents. In this research ACE...

The protective effect of Sambucus ebulus against lung toxicity induced by gamma irradiation in mice

The aim of present study was to investigate the potential antioxidant and lung protective activities of Sambucus ebulus (SE) against toxicity induced by gamma irradiation. Hydroalcoholic extract of SE (20, 50 and 100 mg/kg) was studied for its lung protective activity. Phenol and flavonoid contents of SE were determined. Male C57 mice were divided into ten groups with five mice per group. Only ...
